d'Adda di Fagagna studied Biology at the University of Trieste and under the direction of Arturo Falaschi and Mauro Giacca and obtained his PhD at SISSA, the International School for Advanced Studies, Trieste. He carries out his postdoc at the Gurdon Institute at the University of Cambridge in the laboratory directed by Steve Jackson, studying the response of cells to DNA damage, the DDR. Here d'Adda di Fagagna discovers that DDR activation is in fact the molecular mechanism responsible, for the proliferative arrest characteristic of cellular senescence, under normal conditions. Since 2003 he collaborates with Sofia Francia with who characterized how DDR modulates the activity of telomerase (Francia et al. Curr. Biol., 2006; Francia et al., Cell Div. 2007) and in 2012 demonstrated for the first time the existence of DDRNAs and their function in the activation of the DDR.
